Scans confirmed the extent of the damage on Tuesday and Kos has consulted with club doctors about the rehabilitation process.
The Irish recruit suffered the season-ending injury in a tackle, after offloading the ball to Taylah Levy who went on to score and put the GIANTS back within touching distance of the Swans in the third quarter.
Forward Georgia Garnett also sustained a minor injury to her elbow, coming from the field in the second quarter, but returned to play out her 50th match.
Garnett will be a test to take on the Eagles this week depending on her movement through training.
Fellow attacker Alicia Eva also shapes for a potential return to the side and will be a test to play, following four weeks of rehabilitation on her right hamstring.
The former captain has tracked ahead of schedule, after sustaining a minor hamstring injury in the round three loss to the Crows.
Alyce Parker will consult with her surgeon next week, to provide an update on the rehabilitation of her foot injury.
The team head to Perth with plenty of belief, following back-to-back wins over the Bulldogs and Swans.
The GIANTS will name their team to face the Eagles at gwsgiants.com.au, at 6:20pm on Friday.
